Emergency services attended Melbourne Road about 6.20pm on 31 October. Police said the motorcycle lost control and struck a tree.
One occupant, a 26-year-old man from Nunawading, died at the scene. The second occupant, a 29-year-old Blairgowrie man, was airlifted to hospital with life-threatening injuries.
Victoria Police has not specified which man was riding the motorcycle.
Police investigate Blairgowrie motorcycle crash
Detectives from the Major Collision Investigation Unit are investigating the circumstances of the collision. Police have not released a cause.
Victoria Police warned that traffic disruptions were expected in the area following the crash.
Crash occurs during long-weekend road safety operation
The fatal collision occurred during Operation Furlong, a six-day statewide road safety operation running from 30 October to 4 November.
The operation targets speeding, impaired driving, fatigue and mobile phone use. Police said speeding had contributed to 29 per cent of Victorian road fatalities in 2026, based on data current to 11 October.
Police have not identified any of those factors as contributing to the Blairgowrie crash.
Earlier this year, STPL News reported on a serious motorcycle collision in Langwarrin that left a teenager with life-threatening injuries.
